UHPLC/MS/MS System


The UHPLC/MS/MS system is composed of an Agilent 1290 Infinity Ultra High Performance Liquid Chromatograph (UHPLC) coupled with an AB SCIEX QTRAP® 5500 hybrid linear ion-trap triple quadrupole mass spectrometer.

Technology

The UHPLC/MS/MS system is a novel, state-of-the-art analytical instrumentation, which offers the latest technology in liquid chromatography and mass spectrometry.

The Agilent 1290 Infinity UHPLC (pictured left) has one of the best performance characteristics among the currently available ultra performance liquid chromatography systems. With its broad power range (up to 17,400 psi), high flow and gradient composition accuracy, ultra-low delay volume, and low carry-over (<10 ppm), it delivers the key elements for a reliable and robust LC component of a LC/MS/MS system.

The AB SCIEX QTRAP® 5500 system is the next-generation bench top hybrid triple quadrupole Linear Accelerator™ trap mass spectrometer designed for UHPLC/MS/MS analyses. Equipped with the new eQ™ electronics, and the Qurved LINAC® collision cell, this superior instrument provides a unique combination of Linear AcceleratorTM trap and triple quadrupole functionality, offering ultra-fast scan speeds (up to 20,000 Da/sec), and full MS3 capabilities with the highest sensitivity.


Advancement in Linear Ion Trap (LIT) Technology

By applying the LINAC® collision cell technology to the triple quadrupole linear ion trap, the extraction efficiency, hence the sensitivity, increased by about a 100-fold in all LIT scan modes.

The Enhanced Mass Spectrometry (EMS) feature offers better full scan data by trapping and enriching the ions in the trap. The Enhanced Resolution (ER) function may be used to obtain useful isotopic information with best mass accuracy by dramatically increasing the resolution in the LIT. Or the Enhanced Product Ion (EPI) scan mode may be selected to collect high sensitivity, high resolution data on Product Ions while retaining fragile Precursors, and not losing fragmentation information (no 1/3 low mass cut-off). If you still have a matrix interference with your analyte peak when using MRM, the advantages of the MS/MS/MS (MS3) scans may solve this problem. You can eliminate the interference by obtaining the 2nd generation Product Ions and creating a MS-cubed method for the quantitation of your analytes in complex matrices.

Applications

The UHPLC/MS/MS system is a vital tool in the qualitative and quantitative assays.

For example, during the analysis of in vitro or in vivo samples, such as biological samples (plasma, serum, urine, etc.), metabolites, small molecule pharmaceuticals, peptides, bulk chemical materials, etc., a wide variety of survey scans (MS, precursor ion scans, neutral loss scans and MRM) coupled with MS/MS confirmation via Information Dependent Acquisition (IDA), allows for a broad and effective way of sample detection and quantitation.
